摘要:
目的 探討血漿外泌體中鐵蛋白和轉(zhuǎn)鐵蛋白水平與帕金森病之間的關(guān)系。方法 收集2019年10月至2021年10月就診于福建省立醫(yī)院的60例PD患者,同時納入60例健康對照組,抽取兩組受試者血漿,提取血漿中的外泌體,采用ELISA方法對PD患者和對照組的血漿外泌體中的鐵蛋白和轉(zhuǎn)鐵蛋白受體進行定量分析,分析以上指標(biāo)與帕金森的病程及Hoehn-Yahr(H-Y)分級的相關(guān)性。結(jié)果 PD患者血漿外泌體中鐵蛋白水平較對照組明顯升高[(246.87±139.61)ng/μg vs.(447.97±222.85)ng/μg,P<0.01],血漿外泌體中的轉(zhuǎn)鐵蛋白受體水平與對照組之間差異無統(tǒng)計學(xué)意義[(1 217.18±540.54)ng/μg vs.(1 236.32±389.26)ng/μg, P=0.824]。PD患者血漿外泌體中鐵蛋白及轉(zhuǎn)鐵蛋白受體水平與PD患者的H-Y分級無明顯相關(guān)性(P分別為0.966和0.453)。PD患者的病程與血漿外泌體中的鐵蛋白或TfR水平的相關(guān)性分析,差異均無統(tǒng)計學(xué)意義(P分別為0.263或0.414)。結(jié)論 外周鐵代謝紊亂可能參與了PD發(fā)生,但與疾病進展關(guān)系尚不明確,外泌體中鐵蛋白可能成為帕金森病的外周血生物標(biāo)志物。
Abstract:
Objective To investigate the relationship between the levels of ferritin and transferrin in plasma exosomes and Parkinson's disease.Methods A total of 60 patients with PD in Fujian Provincial Hospital from October 2019 to October 2021 were collected, and 60 healthy controls were enrolled at the same time. The plasma of the two groups was extracted and the exosomes were extracted. The ferritin and transferrin receptors in the plasma exosomes of PD patients and the control group were quantitatively analyzed by ELISA.The correlation between the above indexes,the course of Parkinson's disease and the H-Y stage was analyzed.Results Compared with the control group, the level of ferritin in the plasma exosomes of PD patients was significantly higher [(246.87±139.61)ng/μg vs.(447.97±222.85)ng/μg, P<0.01], and there was no significant difference in the level of transferrin receptor between the two groups [(1 217.18±540.54)ng/μg vs.(1 236.32±389.26)ng/μg, P=0.824].There was no significant correlation between the level of ferritin and transferrin receptor in the plasma exosomes and the course of disease or H-Y stage of PD patients(P=0.966 and 0.263, respectively).There was no significant correlation between the duration of PD and the levels of ferritin or TfR in plasma exosomes(P=0.263 and 0.414,respectively).Conclusion The disorder of peripheral iron metabolism may be involved in the occurrence of PD, but the relationship between ferritin and disease progression is not clear. Ferritin in plasma exosomes may be a biomarker of Parkinson's disease.
