摘要:
目的 探討T細(xì)胞活化V結(jié)構(gòu)域Ig抑制因子(VISTA) 和程序性死亡受體-1 (PD-1)信號(hào)通路聯(lián)合阻斷在大劑量單次X射線照射白化小鼠(BALB/C)小鼠雙移植瘤中誘導(dǎo)遠(yuǎn)隔效應(yīng)可能的機(jī)制。方法 構(gòu)建BALB/C小鼠雙移植瘤模型，采用液相懸浮微球蛋白分析技術(shù)檢測(cè)治療前后細(xì)胞因子表達(dá)變化;流式細(xì)胞術(shù)檢測(cè)小鼠移植瘤模型治療前后瘤內(nèi)和脾臟免疫細(xì)胞比例的變化;游標(biāo)卡尺測(cè)量瘤體大小的變化。統(tǒng)計(jì)學(xué)分析以上指標(biāo)。結(jié)果 X射線大劑量照射聯(lián)合VISTA和PD-I拮抗劑(CA170)治療顯著縮小小鼠第1腫瘤和第2腫瘤瘤體，提高了細(xì)胞因子IFN-Y表達(dá)，并增加了腫瘤內(nèi)CD8+T細(xì)胞的浸潤和擴(kuò)增。結(jié)論 大劑量X射線照射聯(lián)合CA170治療可以顯著增強(qiáng)X射線大劑量照射對(duì)小鼠結(jié)腸癌的抑制，并有效地改善腫瘤微環(huán)境。
