摘要:
目的 探討酪氨酸激酶受體1短發(fā)夾RNA（Tie1-shRNA）對(duì)高糖培養(yǎng)的大鼠內(nèi)皮細(xì)胞（rECs）Tie1的表達(dá)的影響。 方法 將rECs分為對(duì)照組、高糖組、高糖+慢病毒組和高糖+Tie1-shRNA組4組。構(gòu)建Tie1-shRNA慢病毒載體，作用于高糖培養(yǎng)的rECs，干擾酪氨酸激酶受體1（Tie1）mRNA的表達(dá)；采用MTT法檢測(cè)細(xì)胞增殖情況，細(xì)胞劃痕實(shí)驗(yàn)檢測(cè)細(xì)胞遷移情況，Western blot檢測(cè)rECs中Tie1、血管內(nèi)皮生長(zhǎng)因子-A（VEGF-A）、基質(zhì)金屬蛋白酶-2（MMP-2）和基質(zhì)金屬蛋白酶-9（MMP-9）的蛋白表達(dá)。 結(jié)果 細(xì)胞增殖活性4組間差異有統(tǒng)計(jì)學(xué)意義（F=48.937，P<0.01），其中高糖組細(xì)胞增殖活性明顯高于對(duì)照組（P<0.01），高糖+Tie1-shRNA組細(xì)胞增殖活性較高糖組和高糖+慢病毒組均明顯減弱（均P<0.01）。細(xì)胞遷移率4組間差異有統(tǒng)計(jì)學(xué)意義（F=26.728，P<0.01），其中高糖組細(xì)胞遷移率比對(duì)照組明顯增加（P<0.01），高糖+Tie1-shRNA組細(xì)胞遷移率較高糖組和高糖+慢病毒組均明顯減弱（均P<0.05）。4組rECs的Tie1、VEGF-A、MMP-2、MMP-9蛋白的表達(dá)差異有統(tǒng)計(jì)學(xué)意義（FTie1=85.902，P<0.01；FVEGF-A=48.937， P<0.01；FMMP-2=169.857，P<0.01；FMMP-9=130.018，P<0.01），其中高糖組rECs的Tie1、VEGF-A、MMP-2、MMP-9蛋白的表達(dá)明顯高于對(duì)照組（均P<0.05），高糖+Tie1-shRNA組rECs的Tie1、VEGF-A、MMP-2、MMP-9蛋白的表達(dá)較高糖組和高糖+慢病毒組均明顯降低（均P<0.05）。 結(jié)論 通過Tie1-shRNA技術(shù)抑制高糖培養(yǎng)的rEC中Tie1的表達(dá)，可下調(diào)VEGF-A、MMP-2、MMP-9蛋白的表達(dá)，減輕高糖誘導(dǎo)的rEC的增殖和遷移。
Abstract:
Objective To investigate the effect of Tie1-shRNA on the expression of tie1 in high glucose cultured rat endothelial cell（rECs）．Methods rECs were divided into four groups：control group, high glucose group, high glucose + lentivirus group and high glucose + Tie1-shRNA group．Tie1-shRNA lentiviral vector was constructed and used to interfere the expression of Tie1 mRNA in high glucose cultured rECs．The viability of rEC was detected by MTT, the migration of rEC was detected by cell scratch tests, and the expressions of Tie1, VEGF-A, MMP2 and MMP-9 were detected by Western blot．Results The difference of cell viability among the four groups was statistically significant (F=48.937, P<0.01).The cell viability of high glucose group was significantly higher than that of control group (P<0.01)．The cell viability of high glucose + Tie1-shRNA group was lower than that of high glucose group and high glucose + lentivirus group (both P<0.01)．The difference of cell migration rate among the four groups was statistically significant (F=26.728, P<0.01)．The cell migration rate of high glucose group was significantly higher than that of control group (P<0.01), and the cell migration rate of high glucose + Tie1-shRNA group was significantly lower than that of high glucose group and high glucose + lentivirus group (both P<0.05)．The differences of the expression of Tie1, VEGF-A, MMP-2 and MMP-9 protein among the four groups were statistically significant (FTie1=85.902, P<0.01; FVEGF-A=48.937, P<0.01; FMMP-2=169.857, P<0.01; FMMP-9=130.018, P<0.01)．The expression of Tie1, VEGF-A, MMP-2 and MMP-9 protein of high glucose group were significantly higher than those of control group (all P<0.05).The expression of Tie1, VEGF-A, MMP-2 and MMP-9 protein in high glucose + Tie1-shRNA group were significantly lower than those in high glucose + lentivirus group and high glucose group (all P<0.05)．Conclusion Tie1-shRNA can decrease the expression of Tie1, VEGF-A, MMP-2 and MMP-9, suppress cell viability and cell migration induced by high glucose.
