摘要:
目的 探討涎腺良惡性疾病與人乳頭瘤病毒(human papilloma virus,HPV)感染的相關(guān)性。方法 采用HPV基因通用引物GP5+/GP6+和CPI/CPIIG應(yīng)用聚合酶鏈反應(yīng)(polymerase chain reaction,PCR)技術(shù)對33例涎腺惡性腫瘤、35例涎腺良性腫瘤及18例涎腺良性非腫瘤病變石蠟包埋組織進(jìn)行HPV基因檢測,PCR產(chǎn)物采用瓊脂糖凝膠電泳與測序進(jìn)行鑒定。通用引物PCR陽性樣本進(jìn)一步采用HPV16/18型特異性引物進(jìn)行PCR檢測。結(jié)果 86例涎腺良惡性病變組織中HPV基因總檢出率為25.6%(22/86),其中惡性腫瘤為18.2%(6/33),良性腫瘤為25.7%(9/35),良性非腫瘤病變?yōu)?8.9%(7/18),三者間比較,差異無統(tǒng)計(jì)學(xué)意義(χ2=2.624,P=0.269)。HPV16和HPV18基因在上述陽性樣本中均未檢出。結(jié)論 3種涎腺良惡性病變組織中均存在HPV感染,但HPV感染可能并非涎腺惡性腫瘤的主要致病因素。
Abstract:
Objective To evaluate the correlation between benign and malignant lesions of salivary gland and HPV infection.Methods Polymerase chain reaction(PCR)with consensus primers GP5+/GP6+ and CPI/CPIIG was used to detect HPV DNA in 86 cases of formalin-fixed, paraffin-embedded salivary gland lesion tissues including 33 cases of malignant tumors, 35 cases of benign tumors and 18 cases of non-neoplastic lesions.Type-specific PCR was further conducted to detect E6 gene of HPV16 and HPV18 in positive tissues of consensus PCR.Results The results showed that the total positive rate of HPV DNA was 25.6%(22/86), and it was 18.2%(6/33), 25.7%(9/35)and 38.9%(7/18)for malignant tumors, benign tumors and non-neoplastic lesions, respectively.There was no statistical significance among these three groups(χ2=2.624,P=0.269). HPV16 and HPV18 genes were not detected in the above positive samples.Conclusion HPV exists in different kinds of salivary gland lesion tissues, but it may not play a crucial role in carcinogenesis of malignant salivary gland tumors.
