摘要:
目的 通過生物信息學的方法分析炭疽毒素受體1(ANTXR1)在胃癌中表達及意義,揭示相應的分子機制。方法 檢索多個公共數據庫中有關ANTXR1表達和臨床數據,分析ANTXR1 在胃癌中表達情況及其與包括預后在內的臨床病理特征的關系,并下載ANTXR1的共表達基因,通過基因本體(GO)富集和京都基因與基因組百科全書(KEGG)通路分析預測ANTXR1參與的生物學過程。結果 ANTXR1 在胃癌組織中的表達異常上調,其表達與胃癌患者的T 分期、腫瘤分期、組織分化以及種族相關; 并且ANTXR1高表達的患者預后差(P均<0.05)。ANTXR1共表達基因主要參與細胞外基質組成、細胞外結構、胚胎器官發(fā)育等過程,并顯著富集于多個致癌信號通路、細胞黏附分子、ECM-受體相互作用等相關通路。結論 ANTXR1高表達與胃癌患者預后不良有關,在胃癌發(fā)生發(fā)展中發(fā)揮著促癌作用,可能是胃癌的一個可靠的診療靶點。
Abstract:
Objective To investigate the expression and significance of ANTXR1 in gastric cancer(GC), and to explore the potential molecular mechanisms. Methods We retrieved the expression and clinical data of ANTXR1 in several public databases, and analyzed the expression of ANTXR1 in GC and its relationship with clinicopathological features including prognosis. We further downloaded the co-expression gene of ANTXR1 and predicted the biological processes involved in ANTXR1 by GO and KEGG analysis. Results Compared to normal tissues, ANTXR1 was significantly upregulated in GC. The expression of ANTXR1 was significantly correlated with T stage, tumor stage, tissue differentiation and race of gastric cancer patients. The prognosis of patients with high ANTXR1 expression was poor. All the above analyses had statistical significance(P<0.05). The co-expression genes of ANTXR1 were mainly involved in extracellular matrix composition, extracellular structure, embryonic organ development. Moreover, these genes were significantly enriched in several carcinogenic signaling pathways, cell adhesion molecules and ECM-receptor interactions. Conclusion The high expression of ANTXR1 is associated with poor prognosis of GC patients, which plays a role in promoting the development of GC. It may be a reliable target for diagnosis and treatment of GC.
