摘要:
目的 探討MicroRNA-26b(miR-26b)和轉(zhuǎn)錄因子叉頭框蛋白O1(FOXO1)對(duì)人顆粒細(xì)胞KGN凋亡的影響,并初步探討其潛在的作用機(jī)制。方法 將人顆粒細(xì)胞KGN作為研究對(duì)象,分為miR-26b過(guò)表達(dá)組及過(guò)表達(dá)對(duì)照組、miR-26b抑制劑組及抑制劑對(duì)照組、FOXO1特異性干擾RNA組(FOXO1-siRNA組)及FOXO1特異性干擾RNA對(duì)照組(FOXO1-siRNA NC組)、miR-26b抑制劑和FOXO1-siRNA共轉(zhuǎn)染組,Real-time PCR檢測(cè)miR-26b轉(zhuǎn)染率、FOXO1轉(zhuǎn)染率,雙熒光素酶報(bào)告基因檢測(cè)miR-26b與FOXO1 3'UTR的靶向關(guān)系,流式細(xì)胞術(shù)檢測(cè)細(xì)胞凋亡情況,Western blot檢測(cè)FOXO1、胰島素生長(zhǎng)因子1受體(IGF-1R)、胰島素受體底物1(IRSl)、胰島素受體底物2(IRS2)、磷脂酰肌醇-3-激酶(PI3K)的蛋白表達(dá)水平。結(jié)果 雙熒光素酶基因檢測(cè)報(bào)告證實(shí)miR-26b與FOXO1存在靶向關(guān)系。流式細(xì)胞術(shù)顯示:miR-26b過(guò)表達(dá)組早期凋亡率為[(4.58±0.59)%],高于過(guò)表達(dá)對(duì)照組[(2.78±0.43)%]和miR-26b抑制劑組[(3.41±0.39)%](P均<0.05); miR-26b抑制劑組與抑制劑對(duì)照組差異無(wú)統(tǒng)計(jì)學(xué)意義(P>0.05); FOXO1-siRNA組顆粒細(xì)胞KGN細(xì)胞早期凋亡率升高,且高于miR-26b過(guò)表達(dá)組(P均<0.01); miR-26b抑制劑和FOXO1-siRNA共轉(zhuǎn)染組的凋亡率高于miR-26b抑制劑組(P<0.05)。蛋白免疫印跡實(shí)驗(yàn)顯示:與FOXO1-siRNA NC組相比,FOXO1-siRNA組中FOXO1和PI3K蛋白表達(dá)水平下降,IGF-1R、IRS1、IRS2蛋白表達(dá)水平增高(P<0.05)。結(jié)論 miR-26b通過(guò)靶向FOXO1調(diào)控人顆粒細(xì)胞KGN的凋亡,可能與IGF-lR/PI3K信號(hào)通路相關(guān)。
