摘要:
目的 探討壓電型機(jī)械敏感離子通道元件2(piezoelectric type mechanosensitive ion channel component 2,PIEZO2)對胰腺癌細(xì)胞發(fā)生發(fā)展的影響。方法 采用免疫組織化學(xué)技術(shù)檢測胰腺癌與癌旁組織的PIEZO2陽性率; 將PIEZO2干擾片段轉(zhuǎn)染到胰腺癌細(xì)胞Panc-1中,通過實(shí)時(shí)熒光定量PCR(qPCR)和蛋白免疫印跡(WB)檢測PIEZO2的轉(zhuǎn)錄水平及蛋白水平。以細(xì)胞計(jì)數(shù)試劑盒-8(Cell Counting Kit-8,CCK8)檢測細(xì)胞增殖情況,transwell法檢測細(xì)胞的遷移侵襲能力。通過Illumina HiSeqTMX Ten進(jìn)行cDNA文庫構(gòu)建并測序,分析干擾PIEZO2后Panc-1細(xì)胞內(nèi)差異基因并通過GO富集分析參與基因富集的相關(guān)通路。結(jié)果 胰腺癌組織中PIEZO2表達(dá)水平高于癌旁組織,且高PIEZO2表達(dá)的胰腺癌患者總生存率較低。敲減PIEZO2基因后,qPCR和WB結(jié)果顯示PIEZO2的表達(dá)量較對照組減低,差異具有統(tǒng)計(jì)學(xué)意義。敲減組的增殖、遷移、侵襲能力均低于對照組,且差異具有統(tǒng)計(jì)學(xué)意義。對兩組進(jìn)行轉(zhuǎn)錄組分析發(fā)現(xiàn),兩組分別獲得15 521,15 325個(gè)基因,與敲減組相比,對照組有3 411個(gè)差異表達(dá)基因(differentially expressed genes,DEGs)上調(diào),2 830個(gè)DEGs下調(diào),GO富集分析發(fā)現(xiàn)DEGs基因主要富集于細(xì)胞內(nèi)解剖結(jié)構(gòu)、細(xì)胞連接和細(xì)胞外基質(zhì)。結(jié)論 敲除PIEZO2可抑制細(xì)胞的增殖、遷移和侵襲,PIEZO2通過調(diào)控細(xì)胞解剖結(jié)構(gòu)、細(xì)胞連接和細(xì)胞外基質(zhì)參與胰腺癌的發(fā)生發(fā)展,靶向PIEZO2將是胰腺癌新的治療策略。
