摘要:
目的 探討陳皮提取物對APP/PS-1雙轉(zhuǎn)基因阿爾茨海默病(AD)小鼠學(xué)習(xí)記憶功能的影響。方法 選用APP/PS-1雙轉(zhuǎn)基因AD小鼠16只,隨機分為模型對照組和供試品組,8只/組,另取8只C57小鼠為正常對照組,正常對照組和模型對照組分別灌胃等量的生理鹽水,供試品組灌胃陳皮提取物的生理鹽水溶解液,灌胃體積為10 mL/kg,1次/d,連續(xù)18 d。第11天到第16天,進行Morris水迷宮的定位航行實驗及空間探索實驗。第17天到第18天,進行5 min避暗實驗。結(jié)果 在Morris水迷宮實驗中,與正常對照組比較,模型對照組定位航行第2天、第3天、第5天的潛伏期增加(P<0.05); 與模型對照組比較,供試品組第5天的潛伏期降低(P<0.05)。空間探索階段,各組的目標(biāo)象限路程百分比、時間百分比及站臺穿越次數(shù)差異無統(tǒng)計學(xué)意義(P>0.05)。在避暗實驗中各組的錯誤潛伏期、錯誤次數(shù)及錯誤率差異均無統(tǒng)計學(xué)意義(P>0.05)。結(jié)論 陳皮提取物對APP/PS-1雙轉(zhuǎn)基因AD小鼠的空間學(xué)習(xí)、記憶能力有一定的改善作用。
Abstract:
Objective To explore the effect of Citri Reticulatae Pericarpium extracts on the learning and memory ability of APP/PS-1 double transgenic Alzheimer's disease(AD)mice.Methods A total of 16 APP/PS-1 double transgenic AD mice were selected.They were randomly divided into model control group and test group(Citri Reticulatae Pericarpium extracts, 986 mg/kg), eight animals per group, another eight C57 mice were selected as the normal control group.Normal control group and model control group were respectively given the same amount of normal saline, the test group was given normal saline solution of Citri Reticulatae Pericarpium extracts.The volume of intragastric administration was 10 mL/kg once a day for 18 consecutive days.From day 11 to day 16, the positioning navigation experiment and space exploration experiment of Morris water maze were carried out.From the 17th to the 18th day, 5 min dark avoidance test was carried on.Results In the Morris water maze experiment, compared with the normal control group, the incubation period of the model control group was increased on the 2nd, 3rd and 5th day of positioning navigation(P<0.05).Compared with the model control group, the incubation period of the test group on day 5 was decreased(P<0.05).During the space exploration phase, there was no statistically significant difference(P>0.05)in the target quadrant distance percentage, time percentage, and platform crossing frequency among the groups.There were no significant differences in error latency, error times and error rate among all groups in the dark avoidance experiment(P>0.05).Conclusion Citri Reticulatae Pericarpium extracts can improve the spatial learning and memory ability of APP/PS-1 double transgenic AD mice.
