摘要:
目的 以天然脂肪酸月桂酸和硬脂酸為骨架,利用它們?cè)诙谆柞0?dimethylformamide,DMF)中的自組裝構(gòu)建了一種脂質(zhì)體納米載體并負(fù)載阿霉素,對(duì)其靶向人骨肉瘤細(xì)胞Saos-2及凋亡情況進(jìn)行考察。方法 采用自組裝制備了負(fù)載阿霉素的脂質(zhì)體納米載體,采用激光共聚焦考察了其靶向人骨肉瘤細(xì)胞Saos-2能力,采用Annexin V-FITC染色技術(shù)考察了脂質(zhì)體納米載體對(duì)人骨肉瘤細(xì)胞Saos-2凋亡的影響。結(jié)果 自組裝脂質(zhì)體納米載體與人骨肉瘤細(xì)胞Saos-2孵育48 h時(shí),熒光強(qiáng)度最高,綠色熒光的自組裝脂質(zhì)體納米載體會(huì)圍繞在骨肉瘤細(xì)胞Saos-2周?chē)?相對(duì)于同劑量DMF的對(duì)照組,自組裝脂質(zhì)體納米載體組的細(xì)胞無(wú)論是早期凋亡還是晚期凋亡的比例均有所增加。結(jié)論 負(fù)載阿霉素的脂質(zhì)體納米載體能準(zhǔn)確地靶向人骨肉瘤細(xì)胞Saos-2,并加速人骨肉瘤細(xì)胞Saos-2的凋亡。
Abstract:
Objective Using natural fatty acids lauric acid and stearic acid as the backbone, a liposome nanocarrier was constructed through their self-assembly in dimethylformamide(DMF)and loaded with doxorubicin.Its targeting ability to human osteosarcoma cells Saos-2 and apoptosis were investigated.Methods Doxorubicin-loaded liposome nanocarriers were prepared by self-assembly.The targeting ability of the nanocarriers to human osteosarcoma cells Saos-2 was examined using confocal laser scanning microscopy.The effect of liposome nanocarriers on the apoptosis of human osteosarcoma cells Saos-2 was investigated using Annexin V-FITC staining technique.Results When the self-assembled liposome nanocarriers were incubated with human osteosarcoma cells Saos-2 for 48 hours, the fluorescence intensity was the highest.The green fluorescent self-assembled liposome nanocarriers surrounded the osteosarcoma cells Saos-2.Compared to the control group with the same dose of DMF, both early and late apoptotic cell proportions increased in the self-assembled liposome nanocarrier group.Conclusion Doxorubicin-loaded liposome nanocarriers can accurately target human osteosarcoma cells Saos-2 and accelerate their apoptosis.
