摘要:
目的 探討非染色體結(jié)構(gòu)維持蛋白復(fù)合體I G亞基(Non-SMC condensin I complex subunit G,NCAPG)對(duì)大腸癌細(xì)胞生長(zhǎng)的作用和對(duì)增殖凋亡的影響。方法 敲減NCAPG的小干擾RNA(si-NCAPG)轉(zhuǎn)染人大腸癌細(xì)胞株HCT116,Western blot檢測(cè)si-NCAPG對(duì)大腸癌細(xì)胞中NCAPG蛋白表達(dá)的影響; 通過(guò)觀察細(xì)胞形態(tài)、細(xì)胞活力評(píng)價(jià)si-NCAPG對(duì)大腸癌細(xì)胞生長(zhǎng)的影響,并篩選效果最好的序列用于后續(xù)實(shí)驗(yàn)。通過(guò)克隆形成、細(xì)胞周期和凋亡、Western blot實(shí)驗(yàn)評(píng)價(jià)si-NCAPG對(duì)大腸癌細(xì)胞生長(zhǎng)、周期和凋亡的影響。結(jié)果 si-NCAPG顯著降低大腸癌細(xì)胞中NCAPG的蛋白表達(dá)(P<0.05),抑制大腸癌細(xì)胞數(shù)目和細(xì)胞活力(P<0.05),使細(xì)胞阻滯在G2/M期(P<0.05),并促進(jìn)細(xì)胞凋亡(P<0.05)。敲減NCAPG降低Cyclin B1、CDK1和Bcl-2蛋白表達(dá)水平和升高BAX的蛋白表達(dá)水平(P<0.05)。結(jié)論 敲減NCAPG通過(guò)阻滯細(xì)胞在G2/M期和促進(jìn)細(xì)胞凋亡,達(dá)到抑制大腸癌細(xì)胞生長(zhǎng)的作用。
Abstract:
Objective To investigate the effect and mechanism of Non-SMC condensin I complex subunit G(NCAPG)on the growth,proliferation and apoptosis of colorectal cancer cells(CRC).Methods siRNA-NCAPG was transfected into human colorectal cancer cell line HCT116 cells.The effect of si-NCAPG on the expression of NCAPG in HCT116 cells was detected by Western blot.The effect of si-NCAPG on the growth of HCT116 cells was evaluated by observing cell morphology and cell viability, and the best sequences were screened for subsequent experiments.The effects of si-NCAPG on the cell growth, cycle and apoptosis of HCT116 cells were evaluated by clonal formation, cell cycle, apoptosis, and western blot.Results si-NCAPG significantly decreased the expression of NCAPG protein level(P<0.05), inhibited the cell number and cell viability(P<0.05), induced cell cycle arrest in G2/M phase(P<0.05)and promoted cell apoptosis in HCT116 cells(P<0.05).Knocking down NCAPG reduced expression levels of Cyclin B1, CDK1 and Bcl-2 protein and increased expression levels of BAX protein.Conclusion si-NCAPG significantly inhibits the growth of CRC by inducing cell cycle arrest at G2/M phase and promoting cell apoptosis.
