摘要:
目的 探討富含谷氨酸的WD重復(fù)序列蛋白1(glutamate-rich WD40 repeat containing 1,GRWD1)蛋白表達(dá)水平對(duì)胃癌MKN-45細(xì)胞增殖和侵襲能力的影響。方法 分別構(gòu)建GRWD1過(guò)表達(dá)載體和設(shè)計(jì)siRNA并轉(zhuǎn)染胃癌MKN-45細(xì)胞,設(shè)立GRWD1過(guò)表達(dá)組及對(duì)照組、siRNA基因沉默組及對(duì)照組。采用CCK-8法測(cè)定各組細(xì)胞增殖差異,采用transwell檢測(cè)GRWD1過(guò)表達(dá)和基因沉默對(duì)胃癌細(xì)胞MKN-45侵襲能力的影響。結(jié)果 siRNA組轉(zhuǎn)染MKN-45細(xì)胞24 h后,細(xì)胞增殖水平和侵襲能力明顯受到抑制(P<0.01),過(guò)表達(dá)組轉(zhuǎn)染MKN-45細(xì)胞24 h后,細(xì)胞增殖水平和侵襲能力明顯增強(qiáng)(P<0.01)。結(jié)論 GRWD1與MKN-45胃癌細(xì)胞增殖和侵襲能力有關(guān),下調(diào)GRWD1可抑制MKN-45細(xì)胞增殖水平和侵襲能力。反之,上調(diào)GRWD1可提高M(jìn)KN-45細(xì)胞增殖水平和侵襲能力。
Abstract:
Objective To investigate the effect of GRWD1(Glutamate-rich WD40 repeat containing 1)on the proliferation and invasion ability of gastric cancer MKN-45 cells.Methods GRWD1 overexpression vector and siRNA were constructed and transfected into gastric cancer MKN-45 cells.GRWD1 overexpression group, siRNA group and control group were established.Cell proliferation levels in each group were measured using the CCK-8, and transwell was used to detect the effects of GRWD1 overexpression and gene silencing on the invasion ability of gastric cancer cell line MKN-45.Results The results showed that after transfection of MKN-45 cells with siRNA for 24 hours, the cell proliferation level and invasion ability were significantly inhibited(P<0.01), while after transfection of MKN-45 cells with overexpression vector for 24 hours, the cell proliferation level and invasion ability were significantly enhanced(P<0.01).Conclusion GRWD1 is related to the proliferation and invasion ability of MKN-45 gastric cancer cells, and downregulation of GRWD1 can inhibit the proliferation level and invasion ability of MKN-45 cells.On the contrary, upregulation of GRWD1 can improve the proliferation level and invasion ability of MKN-45 cells.
