摘要:
目的 研究中性粒細(xì)胞源性外泌體對肺腺癌細(xì)胞遷移和侵襲的影響。方法 采用1 μmol/L全反式維甲酸(ATRA)誘導(dǎo)人早幼粒細(xì)胞白血病NB4細(xì)胞分化成中性粒細(xì)胞,MGG染色分析其形態(tài)學(xué)的改變,流式細(xì)胞技術(shù)檢測其表面標(biāo)志物CD11b及CD18的表達(dá)。采用總外泌體試劑盒分離提取NB4細(xì)胞和中性粒細(xì)胞來源的外泌體,利用透射電鏡技術(shù)鑒定外泌體的形態(tài)與大小。通過細(xì)胞劃痕和Transwell遷移實(shí)驗(yàn)分析NB4細(xì)胞和中性粒細(xì)胞來源的外泌體對A549細(xì)胞遷移能力的影響。采用Transwell侵襲實(shí)驗(yàn)研究NB4細(xì)胞和中性粒細(xì)胞來源的外泌體對A549細(xì)胞侵襲能力的影響。結(jié)果 MGG染色結(jié)果顯示NB4細(xì)胞經(jīng)ATRA誘導(dǎo)后分化成中性粒細(xì)胞,且流式細(xì)胞檢測結(jié)果表明中性粒細(xì)胞表面標(biāo)志物CD11b及CD18表達(dá)明顯升高。電鏡下觀察到NB4細(xì)胞和中性粒細(xì)胞來源的外泌體大小為40~100 nm,形態(tài)呈雙層膜,中空,圓形或橢圓形。細(xì)胞劃痕和Transwell實(shí)驗(yàn)證實(shí),中性粒細(xì)胞源性外泌體比NB4細(xì)胞源性外泌體能更明顯地增強(qiáng)肺腺癌細(xì)胞的遷移和侵襲能力。結(jié)論 中性粒細(xì)胞源性外泌體能明顯促進(jìn)肺腺癌細(xì)胞的遷移和侵襲,參與肺腺癌的發(fā)生發(fā)展。
Abstract:
Objective To investigate the influence of neutrophil-derived exosomes on migration and invasion of lung adenocarcinoma cells.Methods All-trans retinoic acid(ATRA)was used to induce differentiation of human promyelocytic leukemia NB4 cells into neutrophils.The morphologic changes of neutrophils were analyzed by MGG staining, and the expressions of CD11b and CD18 for the neutrophils specific markers were detected by flow cytometry.The total exosome kit was used to isolate and extract exosomes derived from NB4 cells and neutrophils, the morphology and size of exosomes were identified by transmission electron microscopy.The wound healing and Transwell assay were used to investigate the effects of NB4-derived and neutrophil-derived exosomes on the migration and invasion of A549 cells.Results The NB4 cells were differentiated into neutrophils successfully and the expressions of CD11b and CD18 were significantly increased after ATRA induction.The NB4-derived and neutrophil-derived exosomes ranged from 40 nm to 100 nm, and were bilayer, hollow, round or oval.The NB4-derived and neutrophil-derived exosomes could promote the migration and invasion of A549 cells, and the effect of neutrophil-derived exosomes in promoting the migration and invasion of A549 cells was more pronounced.Conclusion Neutrophil-derived exosomes could promote the migration and invasion of lung adenocarcinoma cells significantly, and play a major role in the occurrence and progression of lung adenocarcinoma.
