目的 探究孕期DEHP暴露對(duì)大鼠子代雌鼠下丘腦長(zhǎng)鏈非編碼RNA(lncRNA)表達(dá)的影響,分析并預(yù)測(cè)DEHP影響子代下丘腦-垂體-性腺軸與能量代謝的機(jī)制。方法 大鼠孕0~18 d連續(xù)灌胃染毒,子代雌鼠出生后立刻取下丘腦組織并提取總RNA,使用illumina Hiseq測(cè)序平臺(tái)進(jìn)行測(cè)序,篩選出差異表達(dá)的mRNA和lncRNA進(jìn)行GO與KEGG富集分析。利用實(shí)時(shí)熒光定量PCR檢測(cè)技術(shù)驗(yàn)證染毒組與對(duì)照組的下丘腦細(xì)胞中差異表達(dá)的目的lncRNA。結(jié)果 通過RNA-seq測(cè)序分析,篩查到108條差異表達(dá)的lncRNA和342條差異表達(dá)的mRNA。KEGG分析顯示差異表達(dá)的lncRNA主要富集在雌激素信號(hào)通路、Ras信號(hào)通路信號(hào)轉(zhuǎn)導(dǎo)和PI3K-Akt信號(hào)通路等通路。結(jié)論 孕期DEHP暴露后子代雌鼠下丘腦組織存在lncRNA及mRNA的差異表達(dá),且差異表達(dá)的lncRNA和mRNA功能分析結(jié)果顯示產(chǎn)前DEHP暴露可能對(duì)子代雌鼠下丘腦的性腺激素和能量代謝過程有影響。
Abstract:
Objective To investigate the effect of DEHP uterus exposure on the expression of lncRNA in the hypothalamus of offspring female rats, and to analyze and predict the mechanism by which DEHP affects the hypothalamic pituitary gonadal axis and energy metabolism in offspring.Methods Rats were exposed to DEHP continuously by gavage from 0 to 18 days since pregnant.The hypothalamic tissue of the offspring female rats was immediately taken after birth and total RNA was extracted.The Illumina Hiseq sequencing platform was used for sequencing, and differentially expressed mRNA and lncRNA were screened for GO and KEGG enrichment analysis.Results The results showed that 108 significantly differentially expressed lncRNA and 342 significantly differentially expressed mRNA were identified through RNA-seq sequencing analysis.KEGG analysis showed that differentially expressed lncRNA was mainly enriched in estrogen signaling pathway, Ras signaling pathway signal transduction, and PI3K-Akt signaling pathway.Conclusion There are differentially expressed lncRNA and mRNA in hypothalamic tissue of female offspring after DEHP exposure during pregnancy, and the functional analysis results of differentially expressed lncRNA and mRNA indicate that prenatal DEHP exposure may affect the gonadal hormone and energy metabolism process in hypothalamus of female offspring.
