摘要:
目的 分析HMGB1基因在子宮腺肌病上皮間質(zhì)轉(zhuǎn)化過(guò)程中的表達(dá),并探討其與β-catenin的相關(guān)性,以探究子宮腺肌病的發(fā)病機(jī)制,為該病的臨床治療提供新的理論基礎(chǔ)。方法 選取我院收治的行子宮切除術(shù)的子宮腺肌病患者8例,取其在位內(nèi)膜組織樣本為在位組,取其異位內(nèi)膜組織樣本為異位組,并以同期手術(shù)病理證實(shí)正常子宮內(nèi)膜的子宮肌瘤患者為對(duì)照組,采用qRT-PCR檢測(cè)HMGB1基因的表達(dá)水平; Western blot法檢測(cè)HMGB1、E-cadherin、Vimentin、β-catenin的蛋白表達(dá)水平,并分析表達(dá)的相關(guān)性。 結(jié)果 與對(duì)照組相比,在位組及異位組中HMGB1 mRNA和蛋白、Vimentin蛋白、β-catenin蛋白的表達(dá)水平升高,E-cadherin蛋白表達(dá)水平降低(P<0.05); 與在位組相比,異位組中表達(dá)HMGB1 mRNA和蛋白、Vimentin蛋白的表達(dá)水平增加,E-cadherin蛋白表達(dá)水平下降(P<0.05); 但β-catenin蛋白的表達(dá)與異位組和在位組比較,差異無(wú)統(tǒng)計(jì)學(xué)意義(P>0.05)。在相關(guān)性分析中,子宮腺肌病在位組、異位組的HMGB1蛋白表達(dá)與E-cadherin呈負(fù)相關(guān)(P<0.05),與Vimentin、β-catenin呈正相關(guān)(P<0.05)。結(jié)論 HMGB1基因可促進(jìn)子宮腺肌病上皮間質(zhì)轉(zhuǎn)化的發(fā)生,與激活Wnt/β-catenin信號(hào)通路相關(guān)。
Abstract:
Objective To analyze the function of HMGB1(high mobility group box-1)gene on epithelial mesenchymal transition(EMT)in adenomyosis, and explore its correlation with β-catenin. To study the mechanism of adenomyosis and provide a new theoretical basis for its clinical treatment.Methods Eight patients of adenomyosis underwent hysterectomy in Fuzhou Second Hospital were selected.The eutopic endometrial tissue samples were taken as the eutopic group, and the ectopic endometrial tissue samples were taken as the ectopic group.The normal endometrial tissue samples from 8 cases of hysteromyoma underwent operation confirmed by pathological examination were selected as control group.The mRNA expression of HMGB1 was detected by qRT-PCR.And the protein expression of HMGB1, E-cadherin, Vimentin and β-catenin were detected by Western blot.And the correlation was analyzed by correlation analysis.Results Compared with those in control group, the mRNA expression levels of HMGB1 and the protein expression levels of HMGB1, Vimentin, β-catenin in eutopic group and ectopic group were significantly higher, while the protein expression level of E-cadherin in them was significantly lower(P<0.05).Compared with those in eutopic group, the mRNA and protein expression levels of HMGB1 and the protein expression levels of Vimentin in ectopic group were obviously higher, but the protein expression level of E-cadherin in ectopic group was obviously lower(P<0.05). However the difference of the protein expression levels of β-catenin between eutopic group and ectopic group was not statistically significant(P>0.05).In the study of correlation analysis, the protein expression levels of HMGB1 in eutopic group and ectopic group were positive correlated with those of Vimentin、β-catenin(P<0.05), which were negative correlated with those of E-cadherin(P<0.05).Conclusion HMGB1 maybe promote the process of EMT of adenomyosis, and is correlated with the simulation of the Wnt/β-catenin signaling pathway.
