摘要:
目的 探討暢脈樂膠囊對腦缺血再灌注損傷（MCAO）大鼠腦組織的保護(hù)機制。 方法 采用線栓法制備MCAO大鼠模型，隨機分為3組，暢脈樂組采用灌胃方式給藥，每日1次，連續(xù)給藥7天；假手術(shù)組和模型組給予等劑量的生理鹽水。采用mNSS法進(jìn)行神經(jīng)功能缺陷評分；TTC染色計算梗死面積和HE染色觀察腦損傷情況；采用 Real-Time PCR 法和 Western blot 法檢測mGluR1和GABAB mRNA 蛋白表達(dá)情況。 結(jié)果 與模型組相比，暢脈樂組神經(jīng)功能缺損程度評分顯著降低，梗死面積顯著減少（P<0.05）。暢脈樂組mGluR1 mRNA和蛋白表達(dá)水平明顯降低，GABAB mRNA 和蛋白表達(dá)水平明顯升高（P<0.05）。 結(jié)論 暢脈樂膠囊可治療大鼠腦缺血損傷并調(diào)節(jié)腦組織mGluR1和GABAB蛋白水平。
Abstract:
Objective To investigate the protective mechanism of Changmai Le capsule on MCAO rat brain tissue. Method The Middle cerebral artery occlusion (MCAO) rat model was established by suture occlusion method. The rats were randomly divided into 3 groups. After the modeling was successful, Changmaile group was given CMLC contents by intragastric administration, once a day, for consecutive 7 days. Sham operation group and model group were given equal volume of normal saline. MNss method was used to score neurological deficits. TTC staining was used to calculate infarct volume and HE staining was used to observe brain injury. The mRNA and protein expressions of mGluR1 and GABAB were detected by real-time PCR and Western blot. Results Compared with model group, the score and infarct volume of Changmai Le group were significantly decreased (P<0.05). In addition, the mRNA and protein expression levels of mGluR1 in Changmai Group were significantly decreased, while the mRNA and protein expression levels of GABAB were significantly increased (P<0.05). Conclusion Changmai Le capsule can treat cerebral ischemia injury and regulate the levels of mGluR1 and GABAB protein in the brain of rats.
