摘要:
目的 探討血清血管內(nèi)皮生長因子(VEGF)、腦源性神經(jīng)營養(yǎng)因子(BDNF)表達(dá)水平與老年糖尿病(DM)大鼠認(rèn)知功能障礙相關(guān)性,為老年DM認(rèn)知功能障礙患者早期診斷和治療提供參考依據(jù)。方法 將40只18～20月齡清潔級(jí)老年雄性SD大鼠隨機(jī)分為兩組:對照組(C組)和糖尿病組(T組)。C組(n=10):為老年非DM大鼠; T組(n=30):采用高糖高脂飼料喂養(yǎng)+腹腔內(nèi)注射鏈脲佐菌素方法建立老年DM大鼠模型,根據(jù)成模后高血糖持續(xù)時(shí)間長短將T組進(jìn)一步隨機(jī)分為3個(gè)亞組:T1(1周)、T2(3周)、T3(6周),每組10只。采用Morris水迷宮測試各組大鼠認(rèn)知水平,酶聯(lián)免疫吸附法測定各組大鼠血清VEGF、BDNF表達(dá)水平,應(yīng)用Pearson相關(guān)分析法分析血清VEGF、BDNF表達(dá)水平與老年DM大鼠Morris水迷宮測試指標(biāo)的關(guān)系。結(jié)果 老年糖尿病大鼠血清VEGF、BDNF、120 s內(nèi)目標(biāo)象限停留時(shí)間和穿越原平臺(tái)位置的次數(shù)隨著高血糖持續(xù)時(shí)間的延長而降低,而潛伏期隨著高血糖持續(xù)時(shí)間的延長而延長(P<0.05)。經(jīng)Pearson相關(guān)分析可知,血清VEGF、BDNF表達(dá)水平與潛伏期呈負(fù)相關(guān),而與120 s內(nèi)目標(biāo)象限停留時(shí)間和穿越原平臺(tái)位置的次數(shù)呈正相關(guān)(P<0.05)。 結(jié)論 血清VEGF、BDNF表達(dá)降低水平與老年DM大鼠認(rèn)知功能障礙嚴(yán)重程度正相關(guān),臨床上檢測血清VEGF、BDNF表達(dá)水平對早期診斷和預(yù)防老年DM認(rèn)知功能障礙發(fā)生有重要的參考意義。
Abstract:
Objective To investigate the correlation between serum levels of VEGF and BDNF and cognitive dysfunction in elderly diabetic rats, so as to provide reference for early diagnosis and treatment of elderly DM patients with cognitive dysfunction.Methods Forty 18to 20-months-old clean male SD rats were randomly divided into two groups as control group(group C)and diabetic group(group T).Group C(n=10): Aged rats without diabetes; Group T(n=30): The aged DM rat model was established by feeding a high sugar and fat diet to the rats, combined with the intraperitoneal injection of streptozotocin, according to the duration of hyperglycemia after modeling, group T was further randomly divided into three subgroups: T1(1 week), T2(3 weeks), and T3(6 weeks), with 10 rats in each subgroup.Morris water maze was used to test the cognitive level, enzyme-linked immunosorbent assay was used to determine the serum levels of VEGF and BDNF.Pearson correlation was used to analyze the relationship between the serum levels of VEGF and BDNF and Morris water maze test parameters in elderly diabetic rats.Results The serum levels of VEGF and BDNF, the residence time of target quadrant and the times of crossing the original platform within 120 s decreased with the extension of hyperglycemia duration, while the latency prolonged with the extension of hyperglycemia duration(P<0.05).Pearson correlation analysis showed that the serum levels of VEGF and BDNF were negatively correlated with the latency, but positively correlated with the residence time in the target quadrant and the times of crossing the original platform within 120 s(P<0.05).Conclusion The decreased expression of serum VEGF and BDNF are positively correlated with the severity of cognitive dysfunction in elderly DM rats.Clinically, the detection of the expression levels of serum VEGF and BDNF has important reference significance for early diagnosis and prevention of cognitive impairment in elderly DM patients.
