摘要:
目的 研究長鏈非編碼RNA(LncRNA)肺腺癌轉(zhuǎn)移相關(guān)轉(zhuǎn)錄本1(MALAT1)對肺癌細(xì)胞放射治療敏感性的作用。方法 運用實時熒光定量逆轉(zhuǎn)錄聚合酶鏈反應(yīng)(qRT-PCR)檢測肺癌細(xì)胞A549中MALAT1的表達(dá),應(yīng)用慢病毒在A549細(xì)胞中干擾或者過表達(dá)MALAT1,并通過克隆形成實驗檢測干擾或者過表達(dá)MALAT1后A549細(xì)胞放療敏感性的改變,通過Western blot檢測干擾或者過表達(dá)MALAT1后A549細(xì)胞放療后DNA損傷標(biāo)記物γ-h2ax表達(dá)的改變。結(jié)果 通過慢病毒在A549細(xì)胞中干擾MALAT1后,A549細(xì)胞的放射敏感性增加,放療后DNA損傷標(biāo)記物γ-h2ax的表達(dá)增加。通過慢病毒在A549細(xì)胞中過表達(dá)MALAT1后,A549細(xì)胞的放射敏感性降低,放療后DNA損傷標(biāo)記物γ-h2ax的表達(dá)減少。結(jié)論 調(diào)控長鏈非編碼RNA MALAT1可以影響肺癌細(xì)胞的放射治療敏感性,且有可能成為肺癌放療增敏的新靶點。
Abstract:
Objective To study the effect of long non-coding RNA MALAT1 on radiosensitivity of lung cancer cells.Methods qRT-PCR was used to detect the expression of MALAT1 in lung cancer cells A549, lentivirus was used to knockdown or overexpress MALAT1 in A549 cells, and clone formation experiment was used to detect the changes in radiosensitivity of A549 cells after knockdown or overexpression of MALAT1, Western blot was used to detect the changes in the expression of γ-h2ax, a radiation induced DNA damage marker in A549 cells after knockdown or overexpression of MALAT1.Results After the knockdown of MALAT1 in A549 cells by lentivirus, the radiosensitivity of A549 cells increased, and the expression of DNA damage marker γ-h2ax increased after radiotherapy.After overexpressing MALAT1 in A549 cells by lentivirus, the radiosensitivity of A549 cells was reduced, and the expression of DNA damage marker γ-h2ax after radiotherapy was reduced.Conclusion The regulation of long non-coding RNA MALAT1 can affect the radiosensitivity of lung cancer cells, and it may become a new target for lung cancer radiotherapy sensitization.
