摘要:
目的 探討衣康酸二甲酯對非肥胖性糖尿病小鼠(NOD)胰島炎的影響。方法 利用流式細胞儀檢測衣康酸二甲酯對RAW 264.7巨噬細胞表型的影響,通過ELISA觀察其對RAW 264.7分泌的細胞因子(IL-6、IL-12和TNF-a)的影響,同時將衣康酸二甲酯腹腔注射NOD小鼠,進一步探討其對胰島內部炎癥的影響。結果 衣康酸二甲酯有利于降低巨噬細胞M1/M2比率,使巨噬細胞向M2型轉換,同時降低IL-6、IL-12和TNF-α的分泌,50 mg/kg的衣康酸二甲酯使胰島評分達到了1.115±0.782 1。結論 衣康酸二甲酯通過M1/M2巨噬細胞表型轉化可以緩解NOD小鼠胰島炎。
Abstract:
Objective To investigate the effect of dimethyl itaconate on insulitis in NOD mice.Methods The phenotypic changes of RAW 264.7 macrophages induced by dimethyl itaconate were detected by flow cytometry, and the effect of dimethyl itaconate on the secretion of RAW 264.7 cytokines(IL-6、IL-12 and TNF-α)were observed by ELISA.At the same time, dimethyl itaconate was injected intraperitoneally into NOD mice to further explore its effect on islet inflammation.Results Dimethyl itaconate reduced the M1/M2 ratio of macrophages and converted macrophages to M2 type, and also reduced the secretion of IL-6, IL-12 and TNF-α.The islet score of 50 mg/kg dimethyl itaconate reached 1.115±0.782 1.Conclusion Dimethyl itaconate can alleviate insulitis in NOD mice by phenotypic transformation of M1/M2 macrophages.
