目的 通過(guò)生物信息學(xué)的方法分析炭疽毒素受體1(ANTXR1)在胃癌中表達(dá)及意義,揭示相應(yīng)的分子機(jī)制。方法 檢索多個(gè)公共數(shù)據(jù)庫(kù)中有關(guān)ANTXR1表達(dá)和臨床數(shù)據(jù),分析ANTXR1 在胃癌中表達(dá)情況及其與包括預(yù)后在內(nèi)的臨床病理特征的關(guān)系,并下載ANTXR1的共表達(dá)基因,通過(guò)基因本體(GO)富集和京都基因與基因組百科全書(KEGG)通路分析預(yù)測(cè)ANTXR1參與的生物學(xué)過(guò)程。結(jié)果 ANTXR1 在胃癌組織中的表達(dá)異常上調(diào),其表達(dá)與胃癌患者的T 分期、腫瘤分期、組織分化以及種族相關(guān); 并且ANTXR1高表達(dá)的患者預(yù)后差(P均<0.05)。ANTXR1共表達(dá)基因主要參與細(xì)胞外基質(zhì)組成、細(xì)胞外結(jié)構(gòu)、胚胎器官發(fā)育等過(guò)程,并顯著富集于多個(gè)致癌信號(hào)通路、細(xì)胞黏附分子、ECM-受體相互作用等相關(guān)通路。結(jié)論 ANTXR1高表達(dá)與胃癌患者預(yù)后不良有關(guān),在胃癌發(fā)生發(fā)展中發(fā)揮著促癌作用,可能是胃癌的一個(gè)可靠的診療靶點(diǎn)。
Abstract:
Objective To investigate the expression and significance of ANTXR1 in gastric cancer(GC), and to explore the potential molecular mechanisms. Methods We retrieved the expression and clinical data of ANTXR1 in several public databases, and analyzed the expression of ANTXR1 in GC and its relationship with clinicopathological features including prognosis. We further downloaded the co-expression gene of ANTXR1 and predicted the biological processes involved in ANTXR1 by GO and KEGG analysis. Results Compared to normal tissues, ANTXR1 was significantly upregulated in GC. The expression of ANTXR1 was significantly correlated with T stage, tumor stage, tissue differentiation and race of gastric cancer patients. The prognosis of patients with high ANTXR1 expression was poor. All the above analyses had statistical significance(P<0.05). The co-expression genes of ANTXR1 were mainly involved in extracellular matrix composition, extracellular structure, embryonic organ development. Moreover, these genes were significantly enriched in several carcinogenic signaling pathways, cell adhesion molecules and ECM-receptor interactions. Conclusion The high expression of ANTXR1 is associated with poor prognosis of GC patients, which plays a role in promoting the development of GC. It may be a reliable target for diagnosis and treatment of GC.
