摘要:
目的 探討EGCG-CoPt NPs-Apt納米粒子對(duì)三陰乳腺癌細(xì)胞 MDA-MB-231 增殖及遷移的影響。方法 利用合成的EGCG-CoPt 納米粒子,偶聯(lián)EGCG及核酸適配體AS1411來(lái)構(gòu)建EGCG-CoPt NPs-AS1411納米粒子,分別采用亞甲藍(lán)法及Transwell法觀察其對(duì)三陰乳腺癌細(xì)胞 MDA-MB-231 增殖及遷移的影響,最后采用Western blot檢測(cè) MDA-MB-231細(xì)胞中腫瘤轉(zhuǎn)移相關(guān)因子MMP-2、MMP-9、EGFR和VASP的蛋白表達(dá)水平。結(jié)果 成功構(gòu)建復(fù)合型納米粒子EGCG-CoPt NPs-AS1411,與CoPt NPs組、對(duì)照組及EGCG-CoPt NPs相比,EGCG-CoPt NPs-AS1411能顯著抑制三陰乳腺癌細(xì)胞 MDA-MB-231的增殖、遷移及MMP-9、EGFR和VASP蛋白表達(dá),差異有統(tǒng)計(jì)學(xué)意義(P<0.05)。結(jié)論 EGCG-CoPt NPs-Apt納米粒子能充分利用納米粒子和核酸適配體的雙重靶向作用,進(jìn)一步發(fā)揮EGCG及核酸適配體AS1411抑制乳腺癌細(xì)胞增殖、遷移的能力,抑制三陰乳腺癌細(xì)胞 MDA-MB-231的增殖與轉(zhuǎn)移。
Abstract:
Objective To investigate the effect of EGCG-CoPt NPs-Apt nanoparticles on the proliferation and migration of triple negative breast cancer cell MDA-MB-231.Methods EGCG-CoPt NPs-AS1411 nanoparticles were synthesized by coupling EGCG with aptamer AS1411.The effects of EGCG-CoPt NPs-AS1411 on proliferation and migration of triple negative breast cancer cells MDA-MB-231 were observed by methylene blue method and transwell method respectively.Finally, western blot was used to detect the protein expression of MMP-2, MMP-9, EGFR and VASP in MDA-MB-231 cells.Results The composite nanoparticles EGCG-CoPt NPs-AS1411 was successfully constructed.Compared with CoPt NPs group, control group and EGCG CoPt NPs group, the proliferation and migration of MDA-MB-231 cells as well as the expression of MMP-9, EGFR and VASP proteins were significantly inhibited by EGCG-CoPt NPs-AS1411(P<0.05).Conclusion EGCG-CoPt NPs-AS1411 nanoparticles can make full use of the dual targeting effect of nanoparticles and aptamers, further exert the ability of EGCG and aptamer As1411 to inhibit the proliferation and migration of breast cancer cells, and inhibit the proliferation and metastasis of triple negative breast cancer cells MDA-MB-231.
