摘要:
目的 探討辛伐他汀對(duì)伴2型糖尿病(T2DM)牙周病患者牙周膜成纖維細(xì)胞(PDLF)增殖分化的影響。方法 采用體外酶消化聯(lián)合組織塊法分離培養(yǎng)伴T(mén)2DM牙周病患者PDLF。在培養(yǎng)液中加入不同濃度的辛伐他汀,分為對(duì)照組(0 mol/L)和4個(gè)實(shí)驗(yàn)組(藥物濃度分別為10-8mol/L、10-7mol/L、10-6mol/L、10-5mol/L),檢測(cè)各組PDLF增殖能力和ALP活性。結(jié)果 10-7mol/L的辛伐他汀能促進(jìn)PDLF增殖和ALP活性,與對(duì)照組比較,差異有統(tǒng)計(jì)學(xué)意義(P<0.05)。結(jié)論 10-7mol/L辛伐他汀能夠促進(jìn)伴T(mén)2DM牙周病患者PDLF的增殖分化,激發(fā)PDLF被高糖抑制的生物學(xué)活性,促進(jìn)牙周組織的愈合修復(fù)。
Abstract:
Objective To study the effect of simvastatin on the proliferation and differentiation of periodontal ligament fibroblasts(PDLF)in patients with periodontal disease complicated with T2DM.Methods In vitro enzymatic digestion and tissue mass were used to isolate and culture PDLF from the patients with periodontal disease complicated with T2DM.Different concentrations of simvastatin were added to the culture medium and divided into the control group(0 mol/L)and four experimental groups(10-8mol/L, 10-7mol/L,10-6mol/L and 10-5mol/L)to detect the proliferation ability and alkaline phosphatase(ALP)activity of each group.Results 10-7mol/L concentration of simvastatin could promote the proliferation and ALP activity of PDLF in patients with periodontal disease complicated with T2DM, and the difference was statistically significant compared with the control group(P<0.05).Conclusion 10-7mol/L simvastatin can promote the proliferation and differentiation of PDLF in patients with peridontal disease complicated with T2DM, stimulate the biological activity of PDLF inhibited by high glucose, and promote the healing and repair of periodontal tissues.
