摘要:
目的 探討腫瘤相關(guān)成纖維細胞(CAF)通過FGFR4/FGF19信號軸調(diào)控結(jié)直腸癌生長可能機制。方法 將研究對象分為3組,分別是細胞組、細胞+NF組、細胞+CAF組。繪制各組癌細胞生長曲線,采用ELISA法檢測細胞外FGF19因子濃度變化。 結(jié)果 癌細胞與CAF共培養(yǎng)后,癌細胞生長率明顯提高,與其他組比較,差異有統(tǒng)計學(xué)意義(P<0.05); 細胞外FGF19因子濃度明顯提高,與其他組比較,差異有統(tǒng)計學(xué)意義(P<0.05)。結(jié)論 CAF通過促進結(jié)腸癌細胞自分泌FGF19因子,促進癌細胞分裂生長。
Abstract:
Objective To explore the possible mechanism of cancer associated fibroblast(CAF)regulating the growth of colorectal cancer through the signal axis of fibroblast growth factor 4/fibroblast growth factor 19. Methods The experiment was divided into three groups, including cancer cell group, cancer cell+NF group, cancer cell+CAF group. The growth curve of each group was drawn. The changes of extracellular fibroblast growth factor 19 were detected by ELISA. Results After co-culture of cancer cells with CAF, the growth rate of cancer cells was significantly increased, compared with other groups(P<0.05); the concentration of extracellular fibroblast growth factor 19 was significantly increased, compared with other groups(P<0.05). Conclusion CAF can promote the autocrine of fibroblast growth factor 19 and promote the growth of cancer cells.
