目的 研究肥厚性幽門狹窄大鼠幽門中神經(jīng)生長因子(NGF)及神經(jīng)營養(yǎng)因子酪氨酸激酶受體A(TrkA)、p75神經(jīng)營養(yǎng)因子受體(p75NTR)表達情況,探討NGF及其受體TrkA與p75NTR在先天性肥厚性幽門狹窄起病中的作用。方法 將受孕后的SD大鼠隨機分為模型組與對照組,模型組經(jīng)尾靜脈注射給予N-硝基-左旋精氨酸甲酯(L-NAME)20 mg/(kg·d),對照組經(jīng)尾靜脈注射給予同等劑量的生理鹽水。新生鼠于出生后第1、7、14天與第21天分別稱重,生后第2天麻醉處死,留取幽門組織。HE染色后顯微鏡下測量幽門肌層厚度。實時定量聚合酶鏈反應(RT-PCR)檢測幽門組織中的NGF及其受體TrkA與p75NTR的mRNA表達水平。Western blot檢測幽門組織中的NGF及其受體TrkA與p75NT的蛋白質表達水平。結果 與對照組新生鼠比較,模型組新生鼠體質量增長明顯緩慢,體質量較低。模型組幽門肌明顯肥厚。模型組新生鼠NGF的mRNA及蛋白表達顯著減少; 模型組新生鼠TrkA與p75NTR的mRNA及蛋白表達顯著減低,差異有統(tǒng)計學意義(P<0.05)。結論 幽門組織中TrkA與p75NTR降低及NGF的表達減少可能與先天性肥厚性幽門狹窄的發(fā)病有關。
Abstract:
Objective To study the expression of nerve growth factor(NGF), neurotrophic factor tyrosine kinase receptor A(TrkA)and p75 neurotrophic factor receptor(p75NTR)in the pylorus of mice with hypertrophic pyloric stenosis, and to explore the role of NGF and its receptors TrkA and p75NTR in the onset of congenital hypertrophic portal stenosis. Methods Pregnant SD mice were randomly divided into model group and control group. The model group was given 20 mg/(kg·d)of N-nitro-L-arginine methyl ester(L-NAME)by tail vein injection, while the control group was given the same dose of normal saline through tail vein. The newborn mice were weighed on the 1st, 7th, 14th and 21st day after birth. The mice were killed on the 2nd day after birth. The thickness of pylorus muscle layer was measured under microscope after staining. Real time quantitative polymerase chain reaction(RT-PCR)was used to detect the mRNA expression levels of NGF, TrkA and p75NTR in pyloric tissue. Western blot was used to detect the protein expression of NGF, TrkA and p75NTR. Results Compared with the control group, the body weight of the model group was significantly slower than that of the control group. In the model group, the pyloric muscle was obviously hypertrophic. The expression of NGF mRNA and protein was significantly decreased in the model group, and the mRNA and protein expression of TrkA and p75NTR in the model group were significantly decreased(P<0.05). Conclusion The decrease of TrkA, p75NTR and NGF expression in pyloric tissue may be related to the pathogenesis of CHPS.
