目的 通過將血糖儀和血管內(nèi)皮生長因子(VEGF)適配體傳感器相結(jié)合,構(gòu)建一種便攜式適配體傳感系統(tǒng),并將其用于骨肉瘤標(biāo)志物的檢測。方法 將血糖儀作為檢測器,與適配體傳感器相結(jié)合,開發(fā)出一種便攜式血糖儀聯(lián)合VEGF適配體傳感系統(tǒng),在對其反應(yīng)條件(孵育時(shí)間、反應(yīng)時(shí)間、存儲時(shí)間)進(jìn)行優(yōu)化的基礎(chǔ)上測定了其檢測限。同時(shí),選取10份骨腫瘤患者血清進(jìn)行檢測,通過與傳統(tǒng)的ELISA法進(jìn)行比較,評估其準(zhǔn)確度。結(jié)果 成功構(gòu)建了一種便攜式血糖儀聯(lián)合VEGF適配體傳感系統(tǒng),其最佳孵育時(shí)間60 min、反應(yīng)時(shí)間30 min、存儲時(shí)間7 h,最低檢測限為6.5×10-9mol/L(2.58 μg/kg)。該法用于骨肉瘤患者血清的檢測,與ELISA 法檢測結(jié)果差異無統(tǒng)計(jì)學(xué)意義(t=2.322,P>0.05)。結(jié)論 成功構(gòu)建了一種便攜式血糖儀聯(lián)合VEGF適配體傳感系統(tǒng),并將其運(yùn)用到骨肉瘤患者血清檢測,其檢測限低,準(zhǔn)確性好。
Abstract:
Objective To construct a portable aptamer sensing system by combining a blood glucose meter with a vascular endothelial growth factor(VEGF)aptamer sensor, and use it for the detection of osteosarcoma markers.Methods A portable blood glucose meter combined with a VEGF aptamer sensing system was developed, which was determined on the basis of optimizing its reaction conditions(incubation time, reaction time, storage time).At the same time, ten serum samples from bone tumor patients were selected for testing, and their accuracy was evaluated by comparing with traditional ELISA methods.Results A portable blood glucose meter combined with a VEGF aptamer sensing system was successfully constructed.The optimal incubation time was 60 minutes, the reaction time was 30 minutes, and the storage time was 7 hours.The minimum detection limit was 6.5×10-9mol/L(2.58 μg/kg).This method was used for the detection of serum in patients with osteosarcoma which was not statistically significant difference with ELISA method(t=2.322,P>0.05).Conclusion A portable blood glucose meter combined with a VEGF aptamer sensing system is successfully constructed and applied to detect serum of osteosarcoma patients.Its low detection limits and accuracy was good.
