摘要:
目的 研究慢病毒介導(dǎo)的γ突觸核蛋白(SNCG)基因敲除對(duì)人結(jié)腸癌SW1116細(xì)胞在裸鼠結(jié)腸癌肝轉(zhuǎn)移實(shí)驗(yàn)?zāi)P椭械挠绊憽7椒?前期實(shí)驗(yàn)中,通過(guò)SNCG基因敲除并慢病毒轉(zhuǎn)染,構(gòu)建穩(wěn)定表達(dá)的SNCG敲除實(shí)驗(yàn)組(RNAi組)、轉(zhuǎn)染空質(zhì)粒的空質(zhì)粒對(duì)照組(NC組)。復(fù)蘇基因敲除實(shí)驗(yàn)組(RNAi組)、空質(zhì)粒對(duì)照組(NC組)及空白對(duì)照組(CON組)的野生型SW1116細(xì)胞,保脾法建立裸鼠結(jié)腸癌肝轉(zhuǎn)移模型,免疫組化染色、逆轉(zhuǎn)錄聚合酶鏈反應(yīng)(RT-PCR)、蛋白質(zhì)免疫印跡(Western blot)檢測(cè)SNCG基因的表達(dá)。結(jié)果 1)成功構(gòu)建裸鼠結(jié)腸癌肝轉(zhuǎn)移模型。2)與對(duì)照組比較,RNAi組裸鼠移植瘤生長(zhǎng)緩慢,原位成瘤率及肝轉(zhuǎn)移率均明顯下降。3)免疫組化、RT-PCR及Western blot結(jié)果顯示,RNAi組SNCG mRNA和蛋白表達(dá)均明顯下降。結(jié)論 SNCG基因沉默后,裸鼠脾臟原位移植瘤及轉(zhuǎn)移瘤的數(shù)量變少,體積變小。在體內(nèi),靶向SNCG敲除降低了原位移植瘤及肝臟轉(zhuǎn)移瘤中SNCG基因的mRNA及蛋白表達(dá),表明SNCG基因?qū)Y(jié)腸癌的增生、侵襲起積極推動(dòng)作用。利用RNAi技術(shù)抑制SNCG基因的表達(dá)有望為結(jié)腸癌的基因治療提供新靶點(diǎn)、新手段。
Abstract:
Objective To study the effect of the lentivirus-mediated RNA interference targetingγ-synuclein(SNCG)gene on the growth of human colorectal carcinoma cell SW1116 in vivo of nude mice.Methods In earlier experiment, we first to screen SNCG RNAi effective target genes, then, transfected and packaged by the lentiviral vectors.Recombinant lentiviral vectors respectively carrying the SNCG-RNAi and SNCG-NC were successfully constructed.The SNCG-RNAi(RNAigroup), SNCG-NC(NCgroup), and non-transfected Human colorectal carcinoma SW1116 cells(CON group),were thawed.And then the colon cancer liver metastasis model in nude mice was established.In order to find outthe difference, SNCG gene expression in these tumor issues were detected by Immunohistochemistry assay, RT-PCR and Western blot.Results 1)The colon cancer liver metastasis model in nude mice was successfully established.2)Compared with those in NC group and CON group, the tumor in the RNAi group grew slowly and metastasis was evidentlysuppressed.3)The mRNA and protein expression of SNCG of the tumor in the RNAi group were significantly lower according to theIHC result, RT-PCR and Western blot.Conclusion Recombinant lentivirus vector SNCG-RNAi decreased the growth and quantity of the tumor in the colon cancer liver metastasis model,including the primary and metastatic, and it can obviously inhibit SNCG gene mRNA and protein expression.So it prompt that the expression of SNCG gene promoted colorectal cancer to grow and transfer.SNCG is expected to become a potent target for the treatment of colon cancer.
