摘要:
目的 研究ATP敏感性鉀通道(KATP)開(kāi)放劑對(duì)人氣道平滑肌細(xì)胞(HASMCs)肥大的影響及其可能的作用機(jī)制。方法 用TGF-β1誘導(dǎo)HASMCs肥大,ATP敏感性鉀通道開(kāi)放劑尼可地爾(Nico)進(jìn)行干預(yù),將實(shí)驗(yàn)分成Control組、TGF-β1組、TGF-β1+Nico組、Nico組。顯微鏡下及免疫熒光法觀察細(xì)胞形態(tài)變化及α-SMA表達(dá)情況; Western Blot法測(cè)定細(xì)胞α-SMA蛋白表達(dá)和CaMKⅡ、CREB磷酸化情況。結(jié)果 免疫熒光實(shí)驗(yàn)表明TGF-β1能濃度依賴性地促進(jìn)細(xì)胞肥大和α-SMA表達(dá),Nico能下調(diào)α-SMA的表達(dá),減輕細(xì)胞肥大; Western blot實(shí)驗(yàn)表明TGF-β1能升高α-SMA表達(dá)和CaMKⅡ、CREB磷酸化水平,Nico能抑制α-SMA的表達(dá),降低CaMKⅡ、CREB磷酸化水平。結(jié)論 Nico能減輕 TGF-β1誘導(dǎo)的HASMCs肥大,可能與抑制CaMKⅡ/CREB信號(hào)通路傳導(dǎo)有關(guān)。
Abstract:
Objective To investigate the effects of the ATP sensitive potassium(KATP)channel openers on human airway smooth muscle cells(HASMCs)hypertrophy and the mechanism underlying. Methods The hypertrophy of HASMCs was induced by exposing to TGF-β1 in vitro.With the intervention of KATP channel opener(Nicorandil),the experiment was divided into four groups:control group, TGF-β1 group, TGF-β1+Nico group and Nico group.The cellular morphological changes and the expression of α-SMA were observed by the microscope and immunofluorescence,the protein expression of α-SMA and phosphorylation of CaMKⅡ,CREB were detected by Western blot.Results Nicorandil could suppress TGF-β1-induced HASMCs hypertrophy by reducing the expression of α-SMA and phosphorylation of CaMKⅡ,CREB.Conclusion Nicorandil can inhibit TGF-β1-induced HASMCs hypertrophy by reducing the phosphorylation of CaMKⅡ/CREB signaling pathways.
